The\u00a0atomic weight\u00a0of iron is 56; thus the minimum molecular weight of myoglobin is (56 \u00d7 100)\/0.34 = about 16,500. Direct measurements of the molecular weight of myoglobin\u00a0yield\u00a0the same value. The molecular weight of hemoglobin, however, which also contains 0.34 percent iron, has been found to be 66,000 or 4 \u00d7 16,500; thus hemoglobin contains four atoms of iron.<\/p>\n\n\n\n
The method most frequently used to determine the molecular weight of proteins is ultracentrifugation\u2014i.e., spinning in a\u00a0centrifuge\u00a0at velocities up to about 60,000 revolutions per minute. Centrifugal forces of more than 200,000 times the gravitational force on the surface of Earth are achieved at such velocities. The first ultracentrifuges, built in 1920, were used to determine the molecular weight of proteins. The molecular weights of a large number of proteins have been determined. Most consist of several subunits, the molecular weight of which is usually less than 100,000 and frequently ranges from 20,000 to 30,000. Proteins of very high molecular weights are found among hemocyanins, the copper-containing respiratory proteins of\u00a0invertebrates; some range as high as several million.
